When you download a shirt in .tga format you need to place it in the folder that you want.. In Pro Cycling Manager 2010 go to the folder :
C:\Program Files\Cyanide\Pro Cycling Manager - Season 2010\CM_Textures\cycliste\team
you will see a bunch of 3 letter abbreviations that stand for team. Find the team that you are going to pick and open the folder. there will be a file called XXX_maillot where the XXX is the abbreviation that the folder was called.
Name the file you downloaded to XXX_maillot where the XXX will be the 3 letter code that the folder was called.
You may want to backup the other jersey just incase.
